520 _aAbstract: Leaves of P. americana Mill. Contain significant amount of phenols and flavonoids which exhibit antioxidant and hypouricemic properties. This study investigated the presence of quercetin and other hypouricemic compounds in the active hexane fractions of P. americana.Pulverized P. americana leaves were soaked in 80% ethanol and evaporated via rotary evaporator. The crude ethanolic extract was partitioned between distilled water and hexane (1:6) using a separatory funnel. Two grams of hexane extract was subjected to vacuum liquid chromatography (VLC) for the separation of components based on their polarity. Thin layer chromatography (TLC) was employed to monitor the separation of components of the resulting fractions which yielded two active fractions. F1H (20%H:80%E) and F2H (60%H:40%E), as identified by the First Year Medical Students Section C1 2nd semester SY 2012-2013. High performance liquid chromatography (HPLC) analysis of the fraction showed significant peaks situated around the retention time recorded from the standard quercetin chromatogram, which demonstrates a positive result for the presence of quercetin in the sample. Percent (%) composition and concentrationof quercetin in F1H was 1.2655% and 0.006328mg/ml while 5.27598 % and 0.02638 mg/ml for F2H which clearly shows that F2H has greater % composition and quercetin concentration than F1H. Rf values 0.929,0.357, 0.610, 0.829 obtained from the fractions via TLC correspond to phytochemical compounds commonly found in P. american as in apigenin, luteolin, isorhamnetin, quercetin and rutin, respectively. Quercetin, a flavonol with known hypouricemic activity and flavonoids apigenin, rutin, luteolin, and isorhamnetin concurred to have possible contributions to the hypouricemic activity of P. americana
