520 _aABSTRACT: Eosin, a synthetic dye that is used in routine hematoxylin and eosin (H&E) Staining technique in laboratories, has been reported to have effects on the human health and may potentially cause to the environment. This research aims to use natural products, specially the Dragon Fruit (Hylocereuspolyrhizus), to determine its efficacy as a counterstain. The dragon Fruit Extract will be obtained through a simple extraction and distillation method. Factors such as pH of the fruit, light, temperature and storage conditions will be taken into consideration as to preserve the color of the extract. The preparation of the tissue specimen, particularly the pork chop tissue undergone the routine procedure for tissue processing that uses paraffin and sectioning. The tissue staining procedure will follow the H&E Staining procedure for paraffin embedded sections. Twelve slides were used: nine of which were used to counterstain with Dragon Fruit Extract at different minutes; three other slides were used to counterstain with eosin. A Pathologist will evaluate the stained slides based on a criteria provided which includes: a) the degree of staining, b) the clarity of cellular components, and c) the intensity of the staining. The data collected was presented on a tabular form. Slides that were stained with Dragon fruit extract for 3 minutes and 5 minutes were graded as 3+ which is interpreted that the whole tissue was stained. Slides that were stained for 1 minute were graded 2+ which are interpreted as 50% of the tissue show staining color. In term clarity of the cellular components slides that were stained for 3 and 5 minutes were classified as clear thus, cellular components are visible. Slides that were stained for 1 minute were classified as not clear correlating the cellular components are not visible,. In terms of intensity, slides with 1 minute, 3 minute and 5 minutes were graded as contrasting meaning the stain differentiates cellular components from the rest to the tissue
