Abstract: In the field of microbiology, staining has been used as a common auxiliary technique to enhance and distinguish different structures of certain pathogens. A mordant is any chemical that binds with a certain dye or stain and fastens it in its place, such that the dye remains adhered to the organism. This study aimed not only to focus on the potential of Tamarindus indica extract as a substitute mordant in microbiologic staining but also its cost effectiveness and eco-friendly factor compared to that of the commercially prepared mordants. The methods used in this study were followed thoroughly by the researchers in order to obtain the ideal concentration of the substance, as used for the research. In this case, the researchers performed a certain process on the Tamarindus indica seed skin until such time that it became a powdery substance. Moreover, the preparation of an inoculum was carried out, and the isolates were used in the research the cultured as part of the experimental study. Furthermore, a Soxhlet extractor was used to accurately extract the seeds, thus producing a certain amount of tannin needed to obtain its ideal concentration. The researchers used the Gelatin Test to confirm the presence of tannins in the said seed. Finally, a gram stain will be performed as another part of the experimental study along with the use of a commonly used mordant and the one produced in the experimental phase for research, thus producing a potential alternative. A basic research was applied in order to develop such refined innovation.
