Abstract: Formalin is a widely popular fixative used in histopathology of tissues. Due to its ability for fast fixation, economic viability and easy usage it is generally used for effortless processing and array of histological techniques that can execute post-fixation. Routine histological staining methods done in laboratories are based on the application and usage of formalin-fixed, paraffin-embedded tissues. Despite these benefits that can be obtain with formalin, the health and safety risks associated with formalin use is a concern. Screening out regrettable substitutes, a handful of synthetic options like alcoholic fixatives, fixatives for nucleic acids, non-alcoholic substitutes, and fixatives with <10% of formalin are commercially available. Natural fixatives such as different sugars, brown sugars and muscovado sugar specifically, are evaluated by the researches as a potential alternative to formalin usage. Fixation was carried out a room temperature for a period of time of 24 hours. Followed by conventional processing method and the tissue sections will then be examined under light microscope. The researchers may be able to conclude that 25% brown sugar and 25% muscovado sugar is comparable to formalin in terms of tissue fixation. The researchers compared formalin and brown sugar by averaging the results of the pathologist and showed that formalin (18.33) has the same average with 25% brown sugar (18.33), while formalin and muscovado sugar (17.33). The results and observations clearly show that both types of sugar can fix tissue. However, the researchers recommend conducting a further study using a lower concentration of the sugar syrup because it yields better results compared to higher concentrations. |
