Summary, etc. ABSTRACT: Various studies have stated that employees who are exposed to routinely used clearing agent, Xylene, have a higher risk of having severe Reynaud's phenomenon. Xylene is also categorized as a flammable and toxic substance which requires safety precautions. With this, numerous research studies were conducted to expand the choices of clearing agents with low health risks in order to prevent further exposure to Xylene. This study evaluated Canola oil as a dealcoholization agent in tissue processing. This research adopted an experimental design with a total of 18 samples of pork liver utilized in this study. Nine samples were cleared using Canola Oil while the other nine samples were cleared using the routine clearing agent Xylene as the control. Samples were evaluated based on their Rigidity, Gross Transparency, Gross Shrinkage, and Cellular Architecture as to different time intervals - 30, 60, and 90 minutes. The foundation was the data acquired from the experiment performed in the histopathology laboratory. Canola oil was compared to Xylene (control) and evaluated using the Wilcoxon signed rank test. In summary, the results indicate that at 30, 60, and 90 minutes, there are no significant changes between Canola Oil and Xylene in terms of Rigidity, Gross Transparency, Gross Shrinkage, and Cellular Architecture. Further research with a larger sample size is suggested to enhance the robustness of the conclusive findings regarding the potential utility of Canola Oil in this particular application. Keywords: Canola oil, cellular architecture, clearing agent, gross shrinkage, gross transparency. miscibility, rigidity, xylene
