Summary, etc. Abstract: LIpase (EC 3.1.1.3) is one of the enzymes important in maintaining the homeostasis of the human body. Lipase performs essential roles in the processing of dietary lipids. Example of which is to hydrolyze triglycerides into their component fatty acid and glycerol molecules. Clinical chemistry assays for human lipase use serum as sample and are said to be exclusive to the diagnosis of acute pancreatitis. One method used for measuring the level of lipase in the serum uses a turbidimetric principle. This study has a primary purpose of comparing olive oil and canola oil when used as a substrate in the turbidimetric serum lipase determination. To do this, a comparison of the enzymatic activity of lipase to the two oil substrates was made. This study aims to prove whether canola oil can be used as a possible substrate in the turbidimetric method for serum lipase determination. An experimental research was conducted utilizing the use of human samples were taken from the door-to-door recruitment of healthy individuals ages 18-50 years old without history of any pancreatic liver diseases living around West Fairview, Quezon City. Simple random sampling was conducted through the use of the lottery method. The participants were screened and were given informed consent forms. Results and findings gathered were taken from the human samples that underwent the procedures for the turbidimetric assay for serum lipase determination. The results were then used to compare the enzymatic activity of lipase to the possible alternative substrate, canola oil and to the traditional substrate, olive oil. Paired sample t-test was used to compare the experimental results that were gathered. The results revealed that there is a statistically significant difference between the two oil substrate that were used. Therefore, Canola oil cannot be used as possible alternative substrate for serum lipase determination
